Formulation and Evaluation of Benzoyl Peroxide Injection

ABSTRACT:

Objective: The present research has been undertaken for the formulation and evaluation of Benzoyl Peroxide Injection. It is used for Antiacne activity. Methods: Benzoyl peroxide is used as Antiacne. Methyl paraben, Propyl paraben and benzalkoniumchloride were used as preservatives. Water for injection is used as a vehicle. The following parameters were evaluated such as clarity test, leakage test, chearity test, assay and Sterility test. Results: The clearity test shows clear solution. Drug chearity test shows no chear in the vials. Leakage test shows no leakage in the vials. pH and drug content show the F1 batch was better the other batches. IR confirmed that all functional groups are same as benzoyl Peroxide. Conclusion: It was concluded that F1 batch is the good than the other batches. So F1 batch is good for the Parenteral use.

INTRODUCTION:

Truncal acne vulgaris, defined as acne on the back or chest, is a common finding in dermatological practice. Benzoyl Peroxide is used as Antiacne. It is used mostly in Skin diseases or Disorders. It is soluble in water, methanol, ethanol, DMF and DMSO1,2,3.

The skin is the largest organ of the body, with a total area of about 20 Square feet. The skin protects us from microbes and the elements, helps regulate body temperature, and permits the sensations of touch, heat and cold4.

 

MATERIALS AND METHODS:

Material:

Benzoyl Peroxide (Sai chemicals pvt. Ltd. Solapur), methyl paraben, propyl paraben and Benzalkonium Chloride and water for Injection were taken from laboratory grade.

 

Equipment:

Weighing balance (Shimadzu AY220), Uv Visible Double beam spectrophotometer (Systronic 2201) Ampoule Sealing Machine (Swastika India Enterprises).

 

METHODS:

Preparation of Injection:

·       Accurately weigh 1gm of Benzoyl Peroxide and transfer it into a beaker and add 50ml water. Add Methyl Paraben / Propyl Paraben / Benzalkonium chloride into it.

·       And boil it until the clear solution appears. Make up the Volume upto 100ml and transfer it into the vial.

IR spectrum of benzoyl peroxide was obtained by scanning the drugs in the range of 4000 to 400. Major peak observed were as 844.95 (O-O stretching), 1255.52 (C-O), 1488.40 (Benzene) and 1782.12 (-C(O)-O-O-C(O)-) whose presence resembled the structure of benzoyl peroxide.

 

Drug Content:

To determine the drug content the 10mg of Injection formulations were transferred in 100ml of volumetric flask containing water and It was sonicated for 10 min. The volume was made upto 100ml and filtered. 2ml of above solution was further diluted using water upto 10ml.and determine the absorbance at wavelength 245nm by using Uv Visible Spectroscopy.

 

Determination of pH:

The pH of Injection formulations was determined by using digital pH meter. 2ml of injection was dissolved in 100ml of distilled water and stored for 2 hours. The Measurement of PH was determined.

 

Clarity Test:

The Injection containing batches F1, F2 and F3 are clear.

 

Leakage Test: There is no leakage in any batch.

 

Sterility test:

All batches are sterile and complies as per Indian Pharmacopoeia 2010.

 

Chearity Test: There is no chear in any batch.

CONCLUSION:

The clearity test shows clear solution. Drug chearity test shows no chear in the vials. Leakage test shows no leakage in the vials. pH and drug content show the F1 batch was better the other batches. IR confirmed that all functional groups are same as benzoyl Peroxide.
